A ratiometry-induced successive reusable electrochemical aptasensing platform: Efficient monitoring of aflatoxin B1 in peanut

چکیده
Currently, most reusable aptasensors rely on the reassembly of aptamers to regenerate sensing interface. However, it remains challenging achieve successive aptasensor reusability. Herein, for first time, we demonstrate that a ratiometric strategy endows electrochemical high reusability realize detection aflatoxin B1 (AFB1). The model interface used thionine (THI)-graphene nanocomposite and ferrocene (Fc)-labeled aptamer output current signals (ITHI IFc). For analysis, specific recognition AFB1 by caused strip from electrode, value ITHI/IFc varied linearly with concentration over 0.01−100 ng mL−1 range. This relationship also worked when using single sensor multiple testing but completely failed ITHI or IFc alone. unique was ascribed capable eliminating environmental influence both signals. feasibility proposed validated applying AFB1, which showed excellent accuracy reliability compared official method, i.e., standard high-performance liquid chromatography-fluorescence (HPLC-FL). aptasensing platform can be universal advanced promotes practical applications particularly in field rapid assays.

Optimization of Aflatoxin B1 Aptasensing
Combination of aptamers with DNAzymes attracted intense attention for development of DNA-based biosensors for detection of mycotoxins. In the present study a combination of aflatoxin B1 specific aptamer and HRP- (horseradish peroxidase-) mimicking DNAzyme was optimized for detecting aflatoxin B1. Detecting approach is based on the binding affinity of aflatoxin B1 to its specific aptamer and con...
متن کامل[Proportion of aflatoxin B1 contaminated kernels and its concentration in imported peanut samples].
Moldy and split peanut kernels were separated from peanuts exported from Brazil, Sudan, India and Taiwan by visual inspection. The remaining peanuts from Brazil, Sudan and India were roasted lightly and the skins were removed. Stained peanuts were separated from the others. Aflatoxin was detected in moldy and stained peanuts. There was a positive correlation between % of aflatoxin-contaminated ...
متن کاملDetoxification of Aflatoxin B1 by Zygosaccharomyces rouxii with Solid State Fermentation in Peanut Meal
Aflatoxins are highly carcinogenic, teratogenetic, and morbigenous secondary metabolites of Aspergillus flavus and A. parasiticus that can contaminate multiple staple foods, such as peanut, maize, and tree nuts. In this study, Zygosaccharomyces rouxii was screened out and identified from fermented soy paste-one kind of traditional Chinese food-to detoxify aflatoxin B₁ (AFB₁) by aerobic solid st...
متن کاملAn Electrochemical Impedimetric Aptasensing Platform for Sensitive and Selective Detection of Small Molecules Such as Chloramphenicol
We report on the aptadetection of chloramphenicol (CAP) using electrochemical impedance spectroscopy. The detection principle is based on the changes of the interfacial properties of the electrode after the interaction of the ssDNA aptamers with the target molecules. The electrode surface is partially blocked due to the formation of the aptamer-CAP complex, resulting in an increase of the inter...
متن کاملAssessment of aflatoxin B1 myocardial toxicity in rats: mitochondrial damage and cellular apoptosis in cardiomyocytes induced by aflatoxin B1
Objective The number of deaths from heart disease is increasing worldwide. Aflatoxin B1 (AFB1), a toxin produced by the fungi Aspergillus flavus and Aspergillus parasiticus, is frequently detected in improperly processed/stored human food products. While AFB1 hepatotoxicity and carcinogenic properties have been well addressed, its myocardial toxicity is poorly documented. This study aimed to in...
